Here are some of the more obvious Twilight mistakes:
Continuity: When Charlie and Bella pull up to the house in the beginning, Bella's truck is in the driveway. They go to the trunk of the cruiser to get her bags out, when they close the trunk - the truck is gone. Then Billy and Jacob pull up in the truck.
Visible crew or equipment: Bella and Jacob are in the old truck for the first time. You can see through the passenger window in the background, crew people working with a large white screen and other equipment.
Visible crew or equipment: When Bella pulls into the school parking lot on her first day you can see the boom reflection in her truck window:

Continuity: When Bella pulls into the school parking lot on her first day, Tyler is sitting there with a hoodie on. When he says "Nice ride," he has a vest over his hoodie. The shot pulls out again, and he has no vest on.
Continuity: In the first Biology class when Bella first sits next to Edward two samples are on their shared desk. Edward nudges one towards her and pulls the other towards himself. In the next shot, they're next to each other in the center of the desk.
Continuity: In the diner when Bella and her dad are eating, the label on the A1 bottle faces away from Bella's dad then towards then away in the same scene.
Continuity: When Edward returns in the next Biology class and Bella enters the classroom as she walks over to her desk the large book sitting on the edge repositions itself from sitting horizontally to vertically between shots. The paper above it also moves closer to the edge desk:

Continuity: When Edward saves Bella from being hit by car in the parking lot scene and he pushes the car away, you can see Bella's hair is first inside her coat, then in the next shot the hair is outside her coat:

Revealing errors: When Dr. Cullen treats Bella in the hospital, the x-ray on the upper left behind him is upside down.
Visible crew or equipment: When Bella is wearing a green shirt in her bedroom, you can clearly see the audio wire under her shirt when she lifts her arms up to do something.
Revealing errors Throughout the film, the vampire's faces are paled out, but not their ears and necks. It's especially noticeable in one of the cafeteria scenes where Robert Pattinson's ears are extremely red.
Continuity: Throughout the film, dark roots appear and disappear from Rosalie's hair.
Visible crew or equipment: During the alley/parking lot scene where the young men harass Bella, above them on a warehouse porch is a crew person with a light or a camera. He is there in one scene and then he is gone.
Continuity: When Edward drops Bella off at the police station on the way back from Port Angeles, she doesn't have her bag, which contained the book she picked up from the book store that she uses later on to research the "cold ones". She should have her bag with her because she uses the book that she bought later on in the movie.
Continuity: When Bella confronts Edward in the forest, the flap on his right shoulder of his jacket is unbuttoned, in the next shot its buttoned itself!

Continuity: When Bella confronts Edward in the woods she drops her red backpack in front of a tree. In one of th next shots in the same scene the backpack disappears:

Continuity: When Edward exposes his skin in the sunlight to Bella and when they are laying in the field, Edward has no hair on his chest. However, when Edward and Bella are in Edward's room and are dancing, Edward visibly has chest hair. (In the DVD director's commentary it's stated that this is caused by the CGI covering his hair with sparkles, therefore no hair is visible)
Continuity: In the meadow scene, Edward and Bella are lying very closely together at first. When the camera angle switches, they are suddenly farther apart.
Revealing errors: When Bella goes to meet the Cullens. Esme has a large dark line on her forehead near her hairline which is actually her natural skin showing next to the pale makeup skin.
Visible crew or equipment: When Edward tells Bella "you better hold on tight, spider monkey" you can see his harness pulling his shirt up.
Revealing errors: When Bella is on Edward's back and they are running up the mountain Bella's hair lies flat as if it she were standing. However, if they were going at the speed implied her hair would have been blowing back.
Revealing errors: When Edward is playing the piano for Bella, the sun is shining on his hands, but he isn't sparkling.
Visible crew or equipment: When Edward passes Billy and Jacob Black on the road, a man with a cap and equipment is visible in the window.
Continuity: In the baseball scene when Carlisle hits the ball, you can see Edward and Emmett start to run after it. But, after the close-ups of their faces, they start to run again.
Continuity: When the Cullens are playing baseball, Rosalie slides into home. When she stands up, there is a large stain on the right side of her white pants from the slide. Later, when they are in the garage getting into the various cars to run from James, Rosalie turns away, and her pants are unstained.
Continuity: When leaving from the baseball scene, Emmett's jeep goes from being dirty on the sides with mud from the tires, to being clean.

Continuity: When the Cullens are in the garage preparing to take Bella to safety, Edward closes the trunk lid twice. First time is not so obvious because the camera is on Emmett and Rosalie, but the trunk closes on the right of the screen. Then it shows it closing again with it being the main shot.
Revealing errors: In the ballet studio, James breaks Bella's leg just below the knee. In the prom scene, Bella is wearing a walking brace, intended for ankle or foot injuries. Bella's injury would require a cast or brace that immobilizes her knee.
Continuity: When Bella is in the hospital, her oxygen tubes change location throughout the scene. when it is a close up, the tubes are close to her eyes and when the camera zooms out the tubes are positioned halfway between her eyes and mouth:

Continuity: During the hospital scene, when Edward is "sleeping," his head goes from from leaning on one shoulder to suddenly on the other shoulder in the very next shot:

Audio mistake: In the Prom scene when Bella wonders why Edward will not turn her into a vampire, Edward says, "I'm not going to END your life for you." The words do not match his mouth movement and it appears as though the word "End" has been dubbed over some other word.
Revealing errors: Near the end of the closing credits, a few counties are thanked for their help during filming. Clallam is misspelled "Clallum".
Revealing errors: In the final credits, "Emmett" is misspelled.
Revealing errors: Clair de Lune is incorrectly listed in the credits as "Claire" de Lune.
